This course is a classroom-based course covering all the practical theory about liquid chromatography and its related techniques including liquid chromatography-mass spectrometry. This course is packed full of examples and practical tips, makes full use of props (instrument parts and consumables which are handled and used for demonstration purposes), includes questions throughout the course and a Questions and Answers session at the end of each day making it the course to attend for lots of useful LC knowledge.
The 5-day Complete course is made up of the 3-day Practical Essentials of HPLC & LC-MS course which focuses on the theory, followed by the 2-day HPLC & LC-MS Clinic focusing on method development, troubleshooting and maintenance with an introduction to advanced techniques. Delegates can choose to attend the whole 5 days, the 3 or 2-day course or individual days, to improve their knowledge in a particular area.
The Practical Essentials of HPLC & LC-MS course topics include:
An introduction to HPLC and LC-MS;
Mobile phase including types of solvents used, pumps and plumbing, isocratic and gradient elution.
Sample introduction discussing manual injection and autosamplers.
LC classification looking at how LC systems are classified;
The types of analytical columns used in liquid chromatography are covered in great detail reviewing resolution, selectivity and efficiency, discussing the separation process and chemistry of the stationary phase;
LC detectors discussing both universal and specific detectors covering Ultra Violet/ Visible absorbance (UV-Vis), Fluorescence (FL), Refractive Index (RI) and Photo Diode Array absorbance (PDA or DAD) detectors;
Mass spectrometry detection and the theory, vacuum, atmospheric pressure inlets, various ionisation techniques, mass analysers and tandem mass spectrometry;
Data analysis including qualitative and quantitative analysis techniques;
Sampling preparation techniques including filtration, enrichment & dilution, standards prepared in matrix, Derivatisation, Solid-Phase Extraction (SPE), Solid Phase Micro-Extraction (SPME), Liquid-Liquid Extraction (LLE), QuECHERS and protein precipitation methods.
Calibration Standards (internal and external standards).
During the final 2 days, the The HPLC & LC-MS Clinic course looks at how to choose the HPLC & LC-MS techniques for an application, set-up the instrument, how to develop & optimise methods, the steps in method validation and the benefits of accreditation, and then finally how to maintain & troubleshoot the instrument, techniques and method. Practical method development case studies and troubleshooting chromatograms test the delegate's knowledge.
The course also takes a brief look at advanced techniques to help solve those very tricky applications including detector selectivity and specificity, Mass Spectral Interpretation, UHPLC vs. HPLC (brief introduction to method migration), chromatographic peak deconvolution using tandem mass spectrometry, multi-dimensional LC (LCxLC), HILIC Separations, Ion Pairing Chromatography (IPC), Size Exclusion Chromatography (SEC), Ion Chromatography (IC) and chemometrics.
At the end of the course, delegates will have in-depth knowledge, from the practical point of view, of liquid chromatography and related techniques and a good understanding of the importance of good chromatography, including how to go about method development and validation. They will know how to select the appropriate solvent, sampling technique, sample introduction technique, column and detector or mass spectrometer and then know how to undertake data analysis. They will also know which areas of the instrument require maintenance and the logical steps in troubleshooting.
Please note, this is a classroom-based course only and does not include any hands-on experience in the laboratory.
This course is for those who have a basic knowledge of liquid chromatography and would like to greatly enhance it, learning the practical theory about all available HPLC & LC-MS techniques.
