Sample Preparation for the LC-MS/MS Analysis of Biofluids

Introduction
Participants will learn about the principles and application of multidimensional SPE for highly selective sample clean-up. Properties and performance of tailor-made SPE packing materials (e.g., restricted access materials, molecularly imprinted polymers, mixed-mode materials, etc.), as well as high throughput and miniaturization of SPE-LC will be described in detail. Besides direct injection and on-line SPE of urine and plasma samples, the direct injection and in-line processing of whole blood will be addressed. Also discussed will be the pros and cons of dried blood spot (DBS) sampling and analysis, and a general introduction to the understanding and monitoring of ion suppression/matrix effects in bioanalytical LC-MS/MS will be given. A focus will be on the elimination of ion suppression by an adequate sample pretreatment and separation.
Topics covered include basic goals of sample cleanup, solid-phase extraction, automation strategies, instrumentation, method development, SPE-LC-MS/MS, whole blood processing and application examples.
